Speaker cleaning works because the speaker itself moves. Its diaphragm travels back and forth, pumping air against whatever is blocking it. A microphone has no such driven element — it is a passive sensor. Playing sound at it does not move anything mechanically, so nothing gets expelled. Tools claiming to clean microphones with tones are describing something that does not happen.
Most phones have two or three, in different locations, used in different combinations by different apps. That is why symptoms seem contradictory. Test in this order:

A blocked microphone makes you sound distant, thin or hollow rather than simply quiet. Noise cancellation can make it worse: if the phone is comparing inputs from two microphones and one is obstructed, the processing may misjudge which signal is noise and suppress your voice.
A microphone port is a pinhole covered by extremely fine mesh — considerably more fragile than a speaker grille. A soft dry brush across the surface is the limit. Nothing inserted into the hole. A pin punctures the mesh and can damage the element behind it, converting a cleanable problem into a replacement.
Check that no app has had microphone permission revoked. Check that a case does not cover a mic opening — this is common and easily missed. Restart the device to clear a stuck audio route. And try a different app, since a single misbehaving app can hold the microphone.
Moisture in a microphone port usually resolves as the device dries, over 24 to 48 hours in moving air. Do not attempt to speed it up with heat, and do not insert anything absorbent — paper and cotton shed fibres that stay behind and make things worse.

No. The frequencies used sit inside the range your speaker already reproduces whenever you play music, and the tool runs at moderate volume rather than maximum. The one genuine caution is earbuds: never run ejection tones into in-ear devices while you are wearing them.

Thirty to sixty seconds per cycle, repeated two or three times with short pauses. Running it continuously for many minutes adds nothing once the accessible liquid is out, and only heats the voice coil.
