Most phones have at least two audio outputs that fail independently. Make a call held to your ear (earpiece), then play a voice recording on speaker (main driver). Whichever sounds wrong is the one to work on. If both are equally dull, the cause is more likely a setting than debris.
A soft dry brush worked across the grille lifts surface lint that would otherwise be driven inward by the pressure changes a cycle creates. On an earpiece slot, adhesive putty pressed lightly and lifted straight off works better than brushing, because the residue there is oily rather than loose.
If there is any moisture involved, clear it first and let the device dry. Damp lint compacts into a dense mat that vibration cannot shift, so attacking dust while the grille is wet tends to push the mat deeper rather than out.

Yes. Water is held by surface tension and usually clears in one or two cycles. Dust is wedged mechanically and held by static, so it needs a sweeping range of frequencies and several more passes.
